FDA approved combination pill for Type-2 Diabetes and Hyperlipidemia....:)


Hello readers, here I am going to present recent news about combination pill for Type-2 Diabetes and Hyperlipidemia.

The Food and Drug Administration (FDA) has approved a combination of sitagliptin (Januvia) and simvastatin (Zocor) that treats both type 2 diabetes and hyperlipidemia.


What is the use of this combination drug?
The combination drug Juvisync is designed to help promote drug adherence by simplifying treatment regimens. Type 2 diabetes and hyperlipidemia, both of which increase the risk of cardiovascular disease, are often seen together.
Juvisync-------->Sitagliptin (Januvia)+Simvastatin (Zocor)


So let's discuss about these individual drug profiles:
Sitaglipitin:
Sitagliptin is an antihyperglycemic drug that works by inhibiting dipeptidyl peptidase-4 (DPP4) and promoting the action of endogenous incretin hormones.

So what are incretin hormones?
Incretin hormones are released to maintain normal levels of glucose in response to glucose elevations that occur after a meal. Incretin hormones achieve this by increasing the insulin synthesis and the release of insulin form pancreatic β cells through intracellular signaling pathway. These incretin hormones are metabolized very rapidly by DPP4.

Simvastatin:
Simvastatin lowers lowdensity lipoprotein (LDL) cholesterol by inhibiting HMGCoA reducatase enzyme. This enzyme catalyze the cholesterol biosynthesis. The inhibtion of this enzyme increases the highdensity cholesterol and decrease the LDL cholesterol, total cholesterol and very-low density cholesterol lipo-protein cholesterol and plasma triglycerides.

Statins also work in other ways to decrease the risk of cardiovascular disease events. These include positive effect on vascular endothelium by increasing the bioavailabilty of nitric oxide which promotes the vasodialation. These stattins also promotes reendothelialization and reducing oxidative stress and inhibiting the inflammatory responses. Statins can also stablize the atherosclerotic plaque in blood vessels; ruptured plaque is a central component of acute coronary syndrome. The statins also decrease the thrombotic action in the blood.

Dose combinations for this combo pill:
The does combinations includes the 100mg/10mg, 100mg/20mg and 100mg/40mg of various strength combinations of sitagliptin and simvastatin respectively. The FDA is requiring a postmarketing clinical trial to compare the glucose lowering ability of sitagliptin alone with the sitagliptin in combination with simvastatin because of a potential increase in glucose levels by statins.
